Rodilsya na hutore Kruzhilinom, stanica Veshenskaya, oblast' Voyska Donskogo. V 1920 vstupil v prodovol'stvennyy otryad. Uchastvoval v boyah s belogvardeycami na Donu. V konce 1922 goda priehal v Moskvu. V 1923 godu v gazete "Yunosheskaya pravda" opublikoval pervyyfel'eton "Ispytanie", pervyy rasskaz Sholohova ("Rodinka") poyavilsya v gazete "Molodoy leninec" v 1924 godu. V 1926 vyshel sbornik "Donskie rasskazy" s predisloviem A. S. Serafimovicha (zemlyaka M. Sholohova). V 1924 godu Sholohov vernulsya na Don i poselilsya vstanice Veshenskoy, gde zhil do poslednih dney. Po ego proizvedeniyam postavleny fil'my "Tihiy Don", "Podnyataya celina", "Pastuh", "Sud'ba cheloveka", "Zherebenok", "Nahalenok", "Kogda kazaki plachut", "Donskaya povest'", "V lazorevoy stepi", "Oni srazhalis' za Rodinu" i dr.
V 1939 godu Sholohov stal deystvitel'nym chlenom Akademii nauk SSSR, dvazhdy byl udostoen zvaniya Geroya Socialisticheskogo Truda (1967, 1980). V 1941 godu poluchil Gosudarstvennuyu premiyu SSSR za roman "Tihiy Don". V 1965 godu zaslugi Sholohova peredmirovoy literaturoy byli otmecheny Nobelevskoy premiey.
